OVERALL RESPONSIBILITY
As a member of the Chromatin Biology group within the Chemical Biology Program, establishes and performs primary and secondary screening assays for chromatin modifying enzymes aiming at the development and optimization of specific small molecule probe compounds, with a focus on histone methyltransferases and demethylases. Tasks include obtaining expression constructs, protein purification, evaluating different assay formats, establishing high throughput and high content screens, and cell-based and biochemical secondary assays. Will be a key contact person for collaboration with the BCB/NT platform, working together with the assay development and screening teams and closely collaborating with chemists in the probe development team to enable iterative rounds of compound testing and optimization. Responsible for the delivery of high quality data, works independently to guide projects through assay development, validation, screening, data analysis and review, and probe optimization. Manages communication with the screening and probe optimization project teams, trains and mentors academic researchers (PhD students, postdocs) and guides researchers at all levels to meet project goals and timelines.
CHARACTERISTIC DUTIES
- Responsible for the specification, implementation and validation of assays for high-throughput screening and secondary assays aiming at the development of chemical probes for histone methyltransferases and demethylases. - Works with HTS, automation, Chemical Biology informatics and chemistry staff at all levels to develop, implement, monitor and coordinate all processes. - Works with informatics group to define requirements for capture of assay development protocols and assay data and establish an ELN environment. - Works independently to design and execute research projects; plans, schedules, or coordinates the development of chemical probe compounds for 1-2 histone modifying enzymes. Ensures that critical project requirements, timelines and deliverables are met. - Performs detailed characterization of hits and chemically optimized compound using biochemistry and enzymology based on previously demonstrated success and technical proficiency in these areas. - Determines the feasibility and makes recommendations regarding the acquisition and/or implementation of new technologies, reagents and assay procedures. - Uses demonstrated scientific creativity, collaboration with others and independent thought, to expend technical capabilities and identify new research opportunities. Regularly presents data at internap meetings, participates in external activities, and technical meetings.
SUPERVISION EXERCISED
- Where appropriate, depending on the needs of the project, may supervise scientists, post-docs or technicians. Provides major input to staffing of project teams, and mentors / trains staff.
QUALIFICATIONS
PhD in biochemistry, molecular biology or related field with 3-5 years experience in enzymology, assay development, chemical or chromatin biology. Requires solid experience in the development and utilization of biochemical assays and experience with cell based assays. Experience with high throughput and high content screening a plus. Open mind to consider challenging assay formats, analysis and physiologically relevant cell models. Successful candidate must have a can do attitude, good problem solving skills, communication skills, and organizational skills.
